• Cell Cytosol

Fluorogenic esterase substrates은 살아있는 세포에 무작위로 들어가 esterases에 의해 fluorescent products로 전환됩니다. 이 fluorescent products은 녹색으로 빛나기 때문에 cell viability assay에 사용할 수 있습니다. CFSE는 또한 살아있는 세포막을 통과하는 ester compound입니다. CFSE에서 유래한 fluorescein은 amine-reactive succinimidyl group을 가지고 있기 때문에 세포 내부 또는 세포막에 있는 단백질 또는 기타 amino group에 공유 결합할 수 있습니다. 이 공유 결합된 fluorescein은 안정적이고 몇 주 동안 세포를 추적할 수 있습니다.

• Mitochondria

미토콘드리아는 대부분의 진핵세포에 존재하는 소기관으로, 에너지원으로 ATP를 생성합니다. ATP는 세포의 모든 활동을 지원하는 데 필수적인 에너지 분자이며, 미토콘드리아는 세포당 100~2,000개가 존재합니다. 그 모양과 크기는 세포 유형에 따라 다르고, 미토콘드리아는 전자 전달계를 가지고 있어, 다양한 환원성 염료로 염색할 수 있습니다. MitoRed와 Rh123은 세포막을 쉽게 통과하여 미토콘드리아에 축적되며, 그 형광 강도는 미토콘드리아에서 생성된 ATP의 양을 보여줍니다.

미토콘드리아는 세포의 에너지 공장으로, 세포의 모든 활동을 지원하는 데 필수적인 역할을 하며, 미토콘드리아의 시각화는 미토콘드리아의 기능을 이해하는 데 중요한 역할을 합니다.

• Nucleus

Fluorescent 시약은 핵산과 결합하여 형광을 방출합니다. PI, DAPI, AO, Hoechst 시약과 같은 fluorescent 시약은 이중 나선 DNA 내부에서 중첩됩니다. DAPI 및 Hoechst 시약 분자는 이중 나선 DNA의 작은 홈에 부착됩니다. 반면에 AO는 이중 가닥 DNA 또는 단일 가닥 DNA 및 RNA와 복합체를 형성할 수 있습니다. AO 분자 하나는 이중 나선 DNA의 세 개의 염기쌍과 중첩되어 최대 파장 526nm의 녹색 형광을 방출합니다. AO 분자 하나는 또한 단일 가닥 DNA 또는 RNA의 인산 그룹 하나와 상호 작용하여 최대 파장 650nm의 빨간 형광을 방출하는 aggregated 혹은 stacked 구조를 형성할 수 있습니다. Viable 세포의 세포막은 Hoechst 시약 외에는 이 형광 시약에 투과할 수 없기 때문에 이 시약은 죽은 세포의 형광 지표로 사용할 수 있습니다. Hoechst 시약은 생리학적 조건에서 양전하를 띠고 살아있는 세포막을 통과할 수 있습니다.

• Bacterial Cell

박테리아를 검출하는 방법에는 여러 가지가 있습니다. Agar plate cultivation과 bacteria-specific DNA 증폭이 그 예입니다. CTC를 사용한 형광 염색은 살아있는 박테리아 세포를 검출하는 데 사용되는 또 다른 방법입니다. 이 방법의 장점은 매우 빠른 검출과 VNC(viable but non-culturable) 박테리아 세포를 검출할 수 있다는 것입니다. CTC는 박테리아 세포의 활동에 의해 formazan 시약으로 전환되는 tetrazolium salt입니다. Formazan 시약의 고체 상태는 붉은 형광을 방출합니다. 따라서 살아있는 박테리아 세포는 CTC로 염색할 수 있으며 형광 현미경으로 쉽게 검출할 수 있습니다.
